Peptide Synthesis: Introduction

Peptide synthesis is a known laboratory technique which is used to generate peptides. Peptides are small chains of amino acids that are connected together with the peptide bonds. Peptides play a significant role in several fields, including medicine, biotechnology, and research. They can be used as therapeutic agents, diagnostic tools, or molecular probes for studying biological processes. In peptide synthesis, amino acids are put together in a unique sequence with the help of a step-by-step assembly technique to form the desired peptide. There are two primary methods of performing peptide synthesis: solid-phase peptide synthesis (SPPS) and liquid-phase peptide synthesis (LPPS).

  • Solid-phase peptide synthesis SPPS method is the most commonly and widely used method for peptide synthesis. In this method, first amino acid is anchored to a solid support, which is usually a resin, and building the peptide chain sequentially by adding protected amino acids which is also a part of solid-phase peptide synthesis. The solid phase peptide synthesis involves five steps: activation, coupling, deprotection, repeated coupling and deprotection, final deprotection and cleavage.
  • LPPS is an alternative method that involves solution-phase reactions. It is typically used for synthesizing smaller peptides or peptide fragments. LPPS requires protecting groups to selectively react with specific amino acids and prevent unwanted reactions. In LPPS, the amino acids are activated and sequentially coupled in solution. After each coupling step, the protecting groups are selectively removed, and the process is repeated until the desired peptide sequence is achieved. The final peptide is purified by techniques such as chromatography or crystallization.

Overall, peptide synthesis is a versatile technique that enables the creation of custom-designed peptides with specific amino acid sequences. It allows researchers and scientists to generate peptides for various applications, including drug development, biomarker discovery, and understanding protein structure-function relationships.

Global Peptide Synthesis Market Analysis

The market for peptide synthesis is influenced by several trends, including the increased emphasis on peptide libraries and screening. The rising demand for long and difficult peptides is growing, including cyclic peptides, stapled peptides, and peptide conjugates. These peptides offer unique properties and enhanced stability, making them attractive for drug development and research applications.

Additionally, peptide-based therapeutics are continuously being explored for the treatment of rare genetic diseases as targeting specific peptides can potentially correct or modulate genetic defects. The development of peptide therapies for rare diseases is gaining momentum, which is driven by the advancements in gene editing technologies and personalized medicine approaches. Researchers are increasingly incorporating non-natural amino acids into peptide sequences to enhance their stability, bioactivity, and pharmacokinetic properties. This allows the development of peptide-based therapeutics with improved drug-like characteristics, expanding the potential applications of peptides in drug discovery and delivery. Outsourcing peptide synthesis to contract manufacturing organizations (CMOs) is becoming a popular trend among pharmaceutical and biotech companies, which is leading to higher peptide synthesis market demand. This trend allows companies to focus on their core competencies while accessing high-quality peptides for their drug development pipelines.

Automation and robotics are also being increasingly employed in peptide synthesis processes that enables high-throughput and efficient production. Automated peptide synthesizers and robotic systems facilitate precise control, rapid synthesis, and reduced human error, thereby enhancing the scalability and reproducibility of peptide manufacturing.
