About Vitamin B1

Vitamin B1, also known as thiamine, is a water-soluble vitamin essential for carbohydrate metabolism and proper nerve, muscle, and heart function. Historically, thiamine was first identified in 1897 as "aneurin," later renamed thiamine. It was synthesised in 1936 by Dutch chemists Barend Coenraad Petrus Jansen and Willem Frederik Donath. Today also it plays a crucial role in converting food into energy, supporting the nervous system, and preventing complications such as beriberi. Thiamine is found in various foods, including whole grains, pork, and legumes, and is vital for maintaining overall health.

Properties of Vitamin B1

Vitamin B1 has a molecular weight of approximately 265.25 g/mol and is composed of a pyrimidine and thiazole ring. Thiamine is found in foods at concentrations of about 0.1 to 0.5 mg per 100 grams. The recommended dietary allowance (RDA) for adults is 1.1 mg for women and 1.2 mg for men. Thiamine is easily oxidised in the body to inactive derivatives but can also be phosphorylated to the active cofactor form, thiamine pyrophosphate.

Manufacturing Process of Vitamin B1

The manufacturing process for vitamin B1 (thiamine) begins with the synthesis of amino-propionitrile from 2-cyanoacetamide. This compound is then reacted with methyl formate to produce acrylonitrile, which is subsequently reacted with ammonia. The process continues with the formation of thiamine hydrochloride (HCl) as an intermediate. From thiamine HCl, thiamine mononitrate is synthesised, which is the final product. The process concludes with refining and packaging the vitamin for distribution. This process yields approximately 47.7% of high-purity thiamine in about 3.5 hours.

Synthesis of Vitamin B1 (Thiamine)

Below are the detailed steps involved in the production of vitamin B1:

Step 1: Synthesis of Thiazole Moiety

The first step in the production of thiamine involves the synthesis of the thiazole moiety. This is typically achieved by reacting 2-methyl-4-aminopyrimidine with carbon disulfide and formaldehyde.

Chemical Reaction:

C5H7N3 (2-methyl-4-aminopyrimidine) + CS2 (carbon disulfide) + CH2O (formaldehyde) → C5H4N2S (thiazole moiety) + H2O + H2S

Step 2: Synthesis of Pyrimidine Moiety

The next step is the synthesis of the pyrimidine moiety. This involves the reaction of 2-methyl-4-aminopyrimidine with acetone and sodium ethoxide.

Chemical Reaction:

C5H7N3 (2-methyl-4-aminopyrimidine) + CH3COCH3 (acetone) + C2H5ONa (sodium ethoxide) → C8H10N4O (pyrimidine moiety)

Step 3: Coupling of Thiazole and Pyrimidine Moieties

The final step involves the coupling of the thiazole and pyrimidine moieties to form thiamine. This is achieved through a condensation reaction.

Chemical Reaction:

C5H4N2S (thiazole moiety) + C8H10N4O (pyrimidine moiety) → C12H17N4OS (thiamine)

Applications and Drivers of Vitamin B1

The vitamin B1 (thiamine) market is driven by several key applications, primarily in the pharmaceuticals, animal feed, and food and beverage sectors. In pharmaceuticals, thiamine is crucial for treating conditions like beriberi and Wernicke-Korsakoff syndrome, with its demand fuelled by rising awareness of vitamin deficiencies and the growing prevalence of related health issues such as diabetes and Alzheimer's disease.

The animal feed industry also significantly contributes to the market, as thiamine is essential for livestock health and productivity. Additionally, the food and beverage sector uses thiamine as a fortifying agent in products like bread and cereals, aligning with consumer trends towards health and nutrition. Overall, the increasing health consciousness among consumers, coupled with advancements in drug development and food fortification initiatives, are key drivers of growth in the vitamin B1 market.
